1. Would you nitrate a CBI barrel after you cleaned it But have not shot it yet? (OR) after you have cleaned it, shot it and then cleaned it again?
2. If you have nitrated a barrel before would you do it again?

I think the correct term is nitride or nitriding.
Once a barrel has been nitrided it cant be set back or machined due to the surface hardening.
I would seriously consider who would do the work.

Thanks for the input. I think I will have my next barrel from CBI nitrided when I buy it.

You can rechamber SOME barrels that have had a nitride coating. It depends on who did it. If it was done with a QPQ then it will be very difficult to rechamber. If it is just a normal Q treatment then it will be easy to cut it away. A lot of barrels out there today are not QPQ.
